You paid for a gut health test, collected the sample, and now you are staring at a page full of terms like “alpha diversity,” “calprotectin” and “SCFA profile.” It reads like a report written for someone with a medical degree you never earned.
You do not need a medical degree. Once you know which question each one answers, the report stops being gibberish and starts being useful.
Diversity: how varied is your gut population?
This is usually the headline number, and it comes in two flavours.
Alpha diversity measures variety inside your own gut. How many species are present, and how evenly are they distributed? Think of a cricket squad. A team can carry fifteen players, but if twelve of them are openers, it is not a balanced squad. Diversity is about that balance, not just the count. A gut with fewer species but a sensible spread can be healthier than one with many species where a single strain dominates everything.
Low diversity is one of the more consistent findings in people with digestive complaints, chronic inflammatory conditions, and metabolic problems. What drives it down in India is rarely a mystery. A diet built around polished rice, refined wheat, biscuits, and packaged snacks feeds a narrow set of species, while a diet of varied dals, seasonal sabzis, millets, and fermented foods supports a wider community.
Beta diversity measures how different one gut community is from another. Labs usually use it to compare your sample against a reference group of people of similar age, diet, and region, but the same measure can compare you against your own earlier sample. On its own it is neither good nor bad; it is context, a way of spotting whether your gut is unusual in a healthy way or in a concerning one.

Calprotectin: is your gut inflamed?
Calprotectin is a protein released by white blood cells called neutrophils when they arrive at a site of inflammation. It is essentially a CRP test for the gut. When calprotectin in your stool is low, it makes major active inflammation in your intestinal wall less likely. When it is elevated, something is triggering an immune response there.
Doctors lean on this marker because it helps separate inflammatory bowel disease from irritable bowel syndrome, two conditions that feel similar but are treated very differently. Many assays use around 50 micrograms per gram of stool as the point where a result stops looking clearly normal, and NICE, which evaluates these tests for the UK health service, records that as the most common research cut-off while deliberately leaving the exact value to individual labs to agree locally (NICE, 2013). Clinical pathways often act from 100 upward, and values in the middle band are treated as needing a repeat test rather than a verdict.
The useful part for you is what a normal result does not mean. A normal calprotectin makes active gut-wall inflammation less likely, which genuinely redirects the search toward motility, fermentation patterns, or food triggers. It is not a rule-out on its own: NSAIDs, acid-suppressing drugs, recent gut infections, and age all shift the number, and only a doctor can convert a result into a diagnosis. If you have persistent abdominal symptoms, blood in stool, weight loss, or fever, see a clinician rather than self-clearing on a home result.
Zonulin and the gut barrier
Your intestinal lining is meant to be selective, letting nutrients through while keeping bacteria and undigested particles out. The cells of that lining are joined by tight junctions, and zonulin is one of the proteins that regulates how open those junctions sit.
Picture two guards at a gate holding hands. When zonulin rises, they drop their arms and the gate lets more through than it should. That loosening is what people mean by “leaky gut,” a phrase used so loosely online that it has almost lost meaning, even though increased intestinal permeability is a real, measurable phenomenon.
The honest caveat is stronger than most summaries admit. The widely used commercial “zonulin” test has been shown not to detect zonulin at all — one study identified a different protein, properdin, as the likely assay target (Scheffler, Frontiers in Endocrinology, 2018), and a separate study independently showed the assay does not detect zonulin itself (Ajamin, PLoS ONE, 2019). A home-stool “zonulin” number therefore cannot be interpreted as a measurement of your barrier, and we do not act on it. If barrier health genuinely matters to you, the evidence-backed route is symptoms, medical history, and a clinician, not a single home marker.

Short-chain fatty acids: is your microbiome producing its share?
When bacteria ferment fibre, they produce short-chain fatty acids, mainly butyrate, propionate, and acetate (Koh, Cell, 2016). Each has a distinct job.
Butyrate is the primary fuel for the cells lining your colon. Without enough of it, the lining weakens and inflammation creeps in. It is produced mainly by species like Faecalibacterium prausnitzii and Roseburia when they ferment the fibre in cooked-and-cooled rice, oats, green (unripe) bananas, and legumes.
Propionate travels to the liver and helps regulate glucose and cholesterol production.
Acetate is the most abundant of the three and feeds into appetite and fat metabolism, and some bacteria use it to make butyrate.
A low overall SCFA profile usually points to one of two things: you are not eating enough fermentable fibre to supply the raw material, or the bacterial populations that do the fermenting are missing. The right fix depends entirely on which, which is why SCFA numbers need to be read alongside the composition report rather than on their own.
Indian legume-heavy eating is genuinely good for this. Rajma, chana, moong, and toor dal all provide the kind of fibre your bacteria can ferment into SCFAs. The problem starts when those foods get displaced by white bread, instant noodles, and packaged snacks, which are nearly fermentable-fibre-free.
Secretory IgA: how are your gut defences?
Secretory IgA is an antibody made by immune tissue in your gut and released into the mucus layer lining the intestine. Its job is to coat bacteria so they cannot attach to and push through the gut wall. It works like the neighbourhood watch of your digestive tract, quiet when things are calm, but essential.
Low sIgA suggests your local mucosal defences are running low. Stress is often linked to lower sIgA, though most of that evidence comes from saliva rather than stool, and acute and chronic stress may push it in opposite directions. Faecal sIgA also has no agreed clinical reference range , different commercial labs use different cut-offs , so treat it as a soft signal, not a measurement. That is still worth knowing: if your number sits clearly low alongside a stressful stretch, your gut’s local defences genuinely weaken under prolonged pressure, which is a different thing from being broken.
Very high sIgA can mean the opposite, that the immune system is actively responding to something: an ongoing infection, a parasite, or a repeatedly irritating food protein.
Pancreatic elastase: is your food being digested?
This marker is about your digestive machinery rather than your microbiome. Pancreatic elastase is an enzyme produced by the pancreas and released into the gut to help break down food. Measuring it in stool tells you whether your pancreas is contributing its share of digestive firepower.
Low elastase can mean the pancreas is not contributing its share of digestive enzymes, so food is not fully broken down before it reaches the colon, where the undigested remnants get fermented into gas and discomfort. This is why it sometimes explains a frustrating scenario: eating well and still feeling terrible after meals, with no amount of dietary tinkering helping because digestion itself is impaired.
But it is a screening marker, not a diagnosis. Faecal elastase-1 misses mild and moderate insufficiency, can read falsely low in loose or watery stool , exactly the sample a bloating-and-loose-stools reader will send in , and a genuinely low result needs a doctor’s work-up rather than self-treatment with enzymes.

Putting the report together
No single biomarker is a verdict. Calprotectin flags inflammation but not its cause. Diversity describes the ecosystem but not its output. SCFAs show what the bacteria are producing but nothing about your immune defences.
The value appears when you read them as a set. Low butyrate alongside elevated calprotectin and low sIgA points toward a gut under inflammatory pressure with weakened defences. Normal diversity with low SCFAs points to a feeding problem, a fibre shortfall rather than a microbial one. High elastase with normal inflammation markers points away from the gut entirely and toward diet or motility.
That is the honest way to use a gut test: biomarkers for clues, symptoms for ground truth, and your actual eating and living habits to explain both. If your report feels like a foreign language, buying a bigger report will not help. Have someone read it in the context of your life and turn the numbers into decisions. These patterns are hypotheses to raise with a clinician, not diagnoses in themselves , especially for markers like calprotectin and elastase, which guide medical work-ups rather than replace them.
